Purpose

  • To recover gastrointestinal pathogens, such as Salmonella and Shigella, from clinical specimens[1][2]
  • Inhibits most nonpathogenic enteric organisms

Technical Details

  • The presence of bile salts and dye inhibit the growth of most Gram-positive bacteria
  • Lactose, sucrose, and salicin are carbohydrates in the medium
    • Strains (generally nonpathogenic) that are able to utilize any of these sugars appear as orange-yellow colonies
    • Strains that strictly utilize salicin will appear salmon-pink to orange-yellow
    • Strains unable to utilize any of these and will appear as translucent, light green, or blueish-green (i.e. Salmonella and Shigella)
  • The addition of thiosulfate and ferric ammonium citrate can detect the production of H2S
    • Colonies appear black or have black centers (e.g. Salmonella)[3]

Materials Needed

  • Pre-mixed powder is available for purchase. The components are listed below.[4]
    • Proteose, peptone
    • Yeast extract
    • Bile salts no.3
    • Lactose
    • Saccharose
    • Salicin
    • Sodium chloride
    • Sodium thiosulfate
    • Ferric ammonium citrate
    • Acid fuchsin
    • Bromthymol blue
    • Agar

Notes

  • Salmonella serovar Typhi is a weak H2S producer, so may not appear black
  • Hektoen Agar was named after the institute where studies were conducted[1]
