Purpose

To directly transfer colonies from one plate (referred to as the master plate) to another, enabling the colonies to be tested for growth under multiple conditions without having to streak out the cells individually.[1]

Technical Details

Materials Needed

  • Master plate (this is easier if the candidates have been gridded on the master plate)
  • Recipient plates that will be grown under varying conditions
  • Velvet cloth cut into squares or circle
  • Metal or plastic ring (to attach the velvet cloth firmly to the block)
  • Replicating block (can buy a plastic one, although an upside-down beaker of the appropriate size will work)
