Preparation of Genomic DNA from Bacteria

Purpose

This protocol can be followed to isolate DNA from a given bacteria for further use.

This protocol is based off the protocol found on pages 2.4.1-2.4.2 of Current Protocols in Molecular Biology [1]

Materials Needed

TE Buffer

10% sodium dodecyl sulfate

20 mg/mL proteinase K

5 mL NaCl

CTAB/NaCl solution

24:1 chloroform/isoamyl alcohol

25:24:1 phenol/chloroform/isoamyl alcohol

Isopropanol

70% ethanol
